Summarization and
Application of Structure-Activity Relationship for Phenolic Antioxidants
Zhang Hongyu Abstract The free radical
scavenging activity of phenolic antioxidants is related to the stability of phenoxyl free
radical generated after H-abstraction of the antioxidant.And the structural factors
stabilizing the phenoxyl free radical include: i) Having heteroatom, O, N, S, etc.,
substituted at ortho or para position, and the p-type lone pair orbit of these atoms are
perpendicular to the phenoxyl plane; ii) Having electron-donating groups substituted in
the phenoxyl ring; iii) Forming intramolecular hydrogen bond with the neighboring hydroxyl
group.Employing these structure-activity relationship, the free radical scavenging
activity characters of 10 flavonoid antioxidants have been predicted successfully. 酚类抗氧化剂构效关系的总结与应用 张红雨 寻找高效、无毒、价廉的抗氧化剂一直是食品、医药、化工等领域的研究热点[1-3]。随着研究工作的深入,总结抗氧化剂的构效关系已成为可能。考虑到目前酚类抗氧化剂应用最广泛,而此类抗氧化剂的作用机制主要是通过酚羟基直接清除自由基以中断链式反应,以下将探讨酚类抗氧化剂的结构与清除自由基活性之间的关系,并用实例验证。希望能深化对抗氧化剂的认识,加速抗氧化剂的筛选。
1.2 推电子基团的取代
为了对以上构效关系进行验证与评价, 以下对10种黄酮类抗氧化剂的活性差异进行理论预测, 并与实验结果对照。
