      筛选对铜绿假单胞菌(P. aeruginosa)具有抑菌活性的中药,并基于表面增强拉曼光谱(SERS)技术监测其调控P. aeruginosa生物膜分泌绿脓菌素(PYO)的过程,以系统评估抑菌活性中药对细菌生物膜生长及其毒力因子表达的干预作用。基于筛选得到的黄连、黄芩和金银花三类抑菌中药构建亚抑菌浓度干预的P. aeruginosa生物膜培养模型,采用SERS技术进行光谱检测与定量分析。在亚抑菌浓度范围内,三种抑菌活性中药干预P. aeruginosa生物膜分泌的PYO呈现差异性表达:黄芩对生物膜形成与PYO分泌的抑制效应最显著,金银花对生物膜形成与PYO分泌均有抑制但抑制效应小于黄芩,而黄连在促进生物膜形成的同时却抑制PYO的分泌。本研究基于SERS技术实现了抑菌活性中药干预下P. aeruginosa生物膜分泌PYO的动态监测,为解析抑菌活性中药抑菌作用发挥提供了新的实验依据。

      Screen traditional Chinese medicines with antibacterial activity against P. aeruginosa, and monitor the process of P. aeruginosa secreting pyocyanin under their regulation based on SERS technology, in order to systematically evaluate the intervention effect of antibacterial traditional Chinese medicines on bacterial biofilm growth and the expression of virulence factors.Based on the screened antibacterial Chinese medicines coptis, scutellaria, and honeysuckle, a sub-inhibitory concentration intervention P. aeruginosa biofilm culture model was constructed, and SERS technology was used for spectral detection and quantitative analysis. Within the sub-inhibitory concentration range, three antibacterial active of traditional Chinese medicines showed differential effects on the secretion of pyocyanin by P. aeruginosa biofilms: Scutellaria baicalensis had the most significant inhibitory effect on biofilm formation and pyocyanin secretion, Lonicera japonica inhibited both biofilm formation and pyocyanin secretion but to a lesser extent than Scutellaria baicalensis, while Coptis chinensis promoted biofilm formation but simultaneously inhibited pyocyanin secretion. This study uses SERS technology to achieve dynamic monitoring of pyocyanin secretion in P. aeruginosa biofilms under the intervention of antibacterial active traditional Chinese medicine, providing new experimental evidence for elucidating the antibacterial effects of antibacterial active traditional Chinese medicine.
